Merging RGB tif files, CYMK files and multiple color files to produce multi color images of...

Hello Everyone!

I’m a medical student doing research in glaucoma and have been imaging a lot of retinas using three different filters (Red, Green and Blue). I’m in dire need of writing a script that will automate the process of merging 3 (or more) images into one that displays all colors. Hopefully some sympathetic soul will be able to help me with this problem:

I currently take three images of the same area in retinas. The filters that I use are labeled FITC, DAPI, TexasRed. They correspond to the Green, Blue and Red images that I plan to merge. These colors correspond to molecules that are labeled within the individual retinas. My idea for the ideal script would include the following characteristics:

1.     The script would ask to open the input folder (it will be located on the Desktop to avoid searching through multiple folders) containing original RGB TIFF files and would create a new output file where only the Merged TIFF files would be saved after creating them.

2.     The script would search for files in the input folder that have identical names except for the filter name (blue, green, red), and merge them into a single file. For example:

Patient1_Shiley_0004_ …_red.tif

Patient1_Shiley_0004_ …_green.tif

Patient1_Shiley_0004_ …_blue.tif

Patient1_Shiley_333_ …_red.tif

Patient1_Shiley_333_ …_green.tif

Patient1_Shiley_333_ …_blue.tif

Would produce in the output folder the following merged files:

Patient1_Shiley_0004_ …_merged.tif

Patient1_Shiley_333_ …_merged.tif

The filename text preceding ”_red” or “_green” or “_blue” or “_yellow” is totally arbitrary and will change according to the changing need of organizing the data but the script has to be smart enough to search for identical groups of files that differ only in the last part of the filename (the color filter used) and merge those only.

3.     In case not all colors are used, the script would assign a black background to the color that is not represented in the input file. In this case below the “blue”  color is missing and so it would be replaced with a black image and only molecules that fluoresced red, or green  would be merged. In this case if the molecules co-localize the resulting spot should look yellow (combo of red and green):

4.     The script should also distinguish the following  combination in the input folder and merge appropriately:

Patientx_Thorton_0005_ …_texred.tif     (red channel)

Patientx_Thorton_0005_ …_fitc.tif           (green channel)

Patientx_Thorton_0005_ …_dapi.tif        (blue channel)

The output file would be in this case:

Patientx_Thorton_0005_ …_merged.tif  

5. Finally, if possible a CMYK version would be great to have as well. Also, does any one know if  8 or more colors can be incorporated (I’m wondering what “lab color” tab is about when given a choice for a new file to be in grayscale, RGB, CYMK, etc). The wavelengths that I would be working with (for color reference) if 8 or more colors can be incorporated in this scheme are (from green range to red range):

525nm

565nm

585nm

605nm

625nm

655nm

705nm

800nm

These are the emission wavelengths of the quantum dot nanocrystals produced by Invitrogen. A theoretical data set, assuming it can be done would look like this:

Oh! I almost forgot. I'm running PHOTOSHOP 10.0.1 CS3 Extended on a 2.4 GHz Intel Core 2 Duo iMac running Mac OS X Version 10.5.7

Moving Adobe Photo Catalogs Off Main HD To External HD 2

I am currently running Adobe Elements 3.0 on my desktop. I've practically maxed my hard drive out with catalogs of photos on this program. Can anyone tell me how to move these catalogs to an external drive (Catalogs intact) so that the program will recognize where the photos are? Each one of these catalogs are created and comprised with photos taken from different folders in the (My Pictures Folder). I am trying to free up space on my internal HD by moving these catalogs intact to the external HD where Adobe Elements will still recognize them and then delete the photos on the internal HD to free up space.

Any Ideas

Working with Photoshop CS5 Extended 3D text in AE CS5 2

Hi all!

I'm fairly new to the AE world and recently I've been learning how PS CS5 and AE CS5 work together. I've run into a problem and I was wondering if someone could help me out?

I've created some 3D text using Photoshop CS5 Extended and imported them into AE CS5 as .psd comps (just like all the tutorials said to do). The text remained 3D in the imported comps and that's great! What I'm trying to do is use the 3D text within another comp and animate them. The problem I'm having is when I do bring the text into another comp they no longer remain in 3D! I've tried using the original .psd comp within the new comp but that didn't work. Then I tried copying the 3D text layer into the other comp and that didn't work either.

The question I have is how can I take the 3D text I created and keep them 3D when using them in another comp? Any information would be greatly appreciated!! Thank you very much!
